The move away from Cape Town Stadium is because the Cape Town Sevens will take place in the Mother City on the same weekend.
This will be the second time the Stormers play a home game at Nelson Mandela Bay Stadium, having also faced Welsh side Dragons in a URC clash there in late 2022.
"Our match against the Dragons RFC two years ago was a fantastic experience and we are looking forward to what should be another special occasion at Nelson Mandela Bay Stadium," Stormers coach John Dobson said in a statement.
"We are expecting a vibrant atmosphere in a world-class stadium, with the added excitement of bringing the Champions Cup to the people of Gqeberha."
The Investec @ChampionsCup will reach a new frontier in Gqeberha when we host @RCTofficiel at the Nelson Mandela Bay Stadium in December.

Stormers Rugby CEO Johan le Roux said it will be a good opportunity to connect with supporters in the Eastern Cape.
"Our team is well-supported wherever they go and we know the people of the Eastern Cape will turn out in their numbers to experience an Investec Champions Cup against a renowned RC Toulon side," he said.
