The Italians, who are without a win in the tournament since victory at Murrayfield in 2015, kicked an early penalty through Paolo Garbisi, but the visitors came to life midway through the first half and a couple of tries from Sam Johnson and Chris Harris seemed to put them in control.
Callum Braley's try brought Italy right back into the game, but Harris gave Scotland some breathing space with his second of the match three minutes before the break as the visitors led 19-10 at the break.
The Scots stretched their advantage eight minutes into the second half when a lovely pass from Finn Russell paved the way for Darcy Graham.
Captain Stuart Hogg then added another try in the 61st minute after great work by Ali Price to set him up.
Harris added a second before the home side ended with something of a flourish with two late tries from debutant replacement Ange Capuozzo putting more respectability on the scoreboard.
