Paul Roos Gymnasium in Stellenbosch was initially scheduled to host the school's rugby event, but the venue has been moved to Hoër Landbouskool Boland in the winelands.
The tournament, which will open SA Rugby's 2023 Youth Weeks programme, will take place from 26 to 29 June.
SA Rugby said in a statement that Paul Roos's Markötter Fields have become unfit to play on after heavy rain in recent weeks.
As a result, the decision was taken to move the Grant Khomo Week to Boland Landbou, which is roughly 40km north of Stellenbosch.
SA Rugby said the venues for the U18 Craven Week (George) and the U16 and U18 Girls Weeks (Johannesburg) were unchanged.
