Cricket South Africa said De Kock, who retired from Test cricket at the end of last year, is "undergoing a graduated return to play programme following a recent illness and his progress is being monitored by the medical team".
A call on his availability for Sunday's ODI, which will be the PinkDay ODI at the Wanderers, will be made before the game.
Rickelton, who has been on duty with the Lions in the 1-Day Cup, joined the ODI squad as cover for Hamza, who withdrew from the squad for personal reasons.
